[68Ga]Ga-DOTA-(1P-PEG4) is a radiopharmaceutical diagnostic probe designed for positron emission tomography (PET) imaging of prostate cancer. It consists of a prostate-specific membrane antigen (PSMA)-targeting monomeric ligand conjugated to a polyethylene glycol (PEG4) spacer and a DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) chelator, which is radiolabeled with the positron-emitting isotope Gallium-68. The probe specifically targets PSMA, a cell-surface protein that is highly overexpressed in prostate cancer cells. The inclusion of the PEG4 chain is intended to optimize the probe's pharmacokinetics by increasing hydrophilicity and potentially improving tumor-to-background ratios. In preclinical studies, this monomeric version serves as a structural baseline to evaluate the advantages of multivalent (dimeric) PSMA tracers in terms of tumor accumulation and renal clearance.
